Determination of Vitamin B12 Using UV-Vis Spectrophotometry
UV-Vis spectrophotometry is a widely used analytical technique for determining the concentration of substances like vitamin B12. This method is particularly effective due to its accuracy and simplicity. Vitamin B12, also known as cyanocobalamin, is a cobalt-containing compound that appears as a dark red crystal. It is essential for various metabolic processes in the body. Traditionally, the absorption coefficient method was employed for measuring vitamin B12 content, but this requires precise instrumentation. The standard curve method, on the other hand, is more reliable and less sensitive to instrument variations. In this experiment, we used the standard curve approach at a wavelength of 361 nm, which is specified by the Chinese Pharmacopoeia for accurate determination. The equipment used included the UV-1800PC UV-Visible Spectrophotometer from China Instrument Co., Ltd. The reagents included vitamin B12 reference substance and tablets from Herrida Pharmaceutical Co., Ltd. The solution preparation involved dissolving the tablets and creating a series of diluted samples for analysis. A linear relationship between concentration and absorbance was established using the standard curve method, showing a strong correlation (R² = 0.9937). Precision tests confirmed the reliability of the method, with an RSD of only 0.7%. Stability tests revealed that the solution remained consistent over 24 hours when stored at refrigeration temperature. Repeatability tests further validated the method's consistency, with an RSD of 1.08%. Finally, the content of vitamin B12 in the tablets was determined to be within the acceptable range of 90% to 110%, confirming the product's quality and stability.
